I have a vanilla XFCE setup I haven’t tweaked at all. Some of my applications are rendering in dark mode, which I find very hard to read (its a vision thing). In particular Calibre, but also Gimp. Is there a configuration flag which tells applications to default to dark mode if they have it? If so where can I find it and how do I turn it off?

AFAIK, any GTK application will use the GTK theme the user defined system-wide.
Though following the release of GNOME 42, there might be theming issues with some GTK applications:
GIMP v2.10 includes 4 themes: Dark, Gray, Light or System theme
To change theme go to - Edit > Preferences > Interface > Theme

I tried changing /etc/xdg/xfce4/panel/default.xml:dark-mode to false in the hope it would make a global change, but it didn’t seem to affect anything I could see.
Calibre does have an environment variable it is supposed to look at: CALIBRE_USE_DARK_PALETTE, but so far it seems to be ignoring it and still loads in dark mode when I set the variable to 0 (in /etc/environment; printenv correctly shows CALIBRE_USE_DARK_PALETTE=0).
